Overview

Carboxyamidotriazole is an orally active small molecule with antineoplastic and antiangiogenic properties. It acts primarily as a cytostatic inhibitor of non-voltage-operated calcium channels and calcium channel–mediated signaling pathways, thereby inhibiting angiogenesis, tumor cell growth, invasion, and metastasis. The drug has been investigated in multiple cancer types including lymphoma, lung cancer (notably non-small cell lung cancer), breast cancer, kidney (renal cell) cancer, and ovarian cancer. Carboxyamidotriazole was initially developed by Beijing Xiehe Bioengineering United Company. Clinical studies have shown that it is generally well tolerated with mostly mild to moderate gastrointestinal side effects; dose-limiting toxicities include reversible cerebellar ataxia and confusion at higher doses[1][2][6][8][9].
