Overview

Carboxymethylpullulan-phenylglycyl-doxorubicin conjugate is an experimental **polymer-drug conjugate** consisting of doxorubicin linked to carboxymethylpullulan through a phenylglycyl spacer. It was reported by **Shionogi** as a macromolecular anticancer conjugate designed for intravenous delivery and preferential retention in solid tumors via the enhanced permeability and retention effect, with subsequent antitumor activity attributable to the doxorubicin payload. In murine models, the conjugate showed improved antitumor efficacy and lower systemic toxicity than free doxorubicin, particularly in solid tumor and metastatic models, but it does not appear to have become a marketed product.
