Overview

CARCIK-CD19 is an allogeneic **cell-based CD19-directed immunotherapy** composed of donor-derived cytokine-induced killer cells genetically modified to express an anti-CD19 chimeric antigen receptor, typically using the non-viral Sleeping Beauty transposon system. The product originated at the University of Milano-Bicocca and has been advanced by Formula Pharmaceuticals for hematologic B-cell malignancies, particularly relapsed or refractory B-cell acute lymphoblastic leukemia after allogeneic hematopoietic stem cell transplantation, with additional development in B-cell non-Hodgkin lymphoma, chronic lymphocytic leukemia, refractory B-cell lymphoma, and Richter's syndrome. Mechanistically, the engineered effector cells recognize **CD19** on malignant B cells and mediate cytotoxic killing through CAR-driven immune activation; the therapy is best categorized as an allogeneic CAR-modified CIK cell therapy rather than a conventional small-molecule or antibody drug.
