Carcinoembryonic antigen (CEA) CAR T-cells are an investigational cell-based immunotherapy modality in which patient-derived T cells are genetically engineered to express chimeric antigen receptors (CARs) that specifically target carcinoembryonic antigen (CEA, also known as CEACAM5 or CD66e). CEA is a tumor-associated antigen expressed at high levels across a broad range of epithelial solid tumors, including colorectal cancer, pancreatic cancer, gastric cancer, lung cancer, and breast cancer. The therapy has been explored in multiple clinical trials (e.g., NCT01212887, NCT02349724, NCT01373047, NCT06006390, NCT06043466), both via systemic administration and local hepatic artery infusion. Early first-generation constructs (such as MFEζ, using the MFE23 scFv fused to CD3ζ) showed limited clinical efficacy due to poor T-cell persistence and transient respiratory toxicity. This is not a single branded drug from one company, but rather a research platform pursued by multiple academic and clinical groups.
