Overview

Cardiotrophin-1 (CT-1) is a cytokine and member of the interleukin (IL)-6 family, which also includes leukemia inhibitory factor (LIF), ciliary neurotrophic factor, IL‑11, and oncostatin M. It is a protein composed of 201 amino acids. CT‑1 was originally identified for its ability to induce cardiac myocyte hypertrophy and has both hypertrophic and cardioprotective properties. Its mechanism involves binding to the LIF receptor β, which then recruits glycoprotein 130 (gp130) to form a signaling complex that activates downstream pathways such as JAK/STAT, ERK/MAPK, PI3K/Akt, and others. These pathways mediate effects including cardiac myocyte survival under hypoxic conditions, inhibition of apoptosis, promotion of neuronal survival/differentiation, regulation of glucose/lipid homeostasis, modulation of circadian metabolic rhythms in adipose tissue, stimulation of fibroblast growth/collagen synthesis in the heart, and potential renoprotective effects in acute kidney injury models[3][5][6][7][8]. The drug has been investigated for myocardial reperfusion injury and acute kidney injury; it holds orphan drug status in both the US and EU[7][9].
