Cardizyme is a catalytic antibody (catabody) developed by Covalent Bioscience for the treatment of systemic age-associated amyloid disease, specifically targeting misfolded transthyretin (misTTR). Unlike traditional monoclonal antibodies that bind targets in a 1:1 ratio, Cardizyme acts as an enzyme to selectively hydrolyze and destroy misfolded TTR aggregates while leaving the functional, properly folded TTR protein intact. This catalytic mechanism allows a single antibody molecule to neutralize multiple amyloid aggregates without inducing an inflammatory response. Preclinical data indicated potential for treating cardiac amyloidosis and spinal stenosis, showing significant removal of amyloid deposits in diseased tissues. However, development has ceased following the closure of Covalent Bioscience.
