CARG-2020 is a genetically engineered oncolytic virus developed using the Artificial Virus for Infectious Diseases and Immuno-Oncology (AVIDIO) platform. It is a membrane-encapsulated, self-amplifying RNA replicon (virus-like vesicle) that delivers three immunomodulators within a single vector: interleukin 12 (IL-12), dominant-negative IL-17 receptor A (dn-IL17RA), and short-hairpin RNAs targeting programmed death ligand 1 (PD-L1). This combination enables broad immune modulation in the tumor microenvironment by activating Th1 immune responses, downregulating PD-L1 expression, inhibiting cancer-promoting inflammation, and preventing T cell exhaustion. Preclinical studies have shown that intratumoral administration of CARG-2020 eradicates large established tumors, prevents recurrence, provides long-term survival benefit in mouse models of ovarian and colorectal cancer, and induces systemic immunity capable of controlling both primary and distal tumors. The drug is being developed primarily for solid tumors including ovarian cancer, colorectal cancer, breast cancer, skin neoplasms, peritoneal neoplasms, and other malignancies[1][2][3][4][5][7].
