CARGO-T cells are an investigational autologous chimeric antigen receptor T cell (CAR-T) therapy targeting the epidermal growth factor receptor variant III (EGFRvIII), a tumor-specific mutation frequently expressed in glioblastoma multiforme (GBM). Developed by University College London, this therapy involves the genetic modification of a patient's own T cells to recognize and eliminate EGFRvIII-positive tumor cells. It is currently being evaluated in a Phase I clinical trial for patients with relapsed or refractory glioblastoma, where it is administered via intravenous infusion, with some protocols exploring intracerebroventricular delivery via an Ommaya reservoir. The trial also investigates the potential synergistic effect of combining CARGO-T cells with the CTLA-4 checkpoint inhibitor ipilimumab to overcome the immunosuppressive tumor microenvironment of the brain.
