Caricotamide is a small molecule used as a **co-substrate** in the investigational chemotherapy combination **Prolarix**, where it is administered with tretazicar (also known as CB1954). The drug combination targets tumors by exploiting the enzyme **NQO2 (quinone oxidoreductase 2)** in tumor cells. Tretazicar serves as a prodrug that is bioactivated by NQO2 in the presence of caricotamide, leading to the formation of a potent **bifunctional DNA cross-linking agent** capable of inducing cytotoxic effects in cancer cells through DNA cross-linking. Prolarix was primarily developed for **liver cancer including hepatocellular carcinoma**, but development was discontinued in phase 2 trials.
