Overview

Carlumab + gemcitabine is an investigational combination therapy studied in advanced solid tumors, notably pancreatic ductal adenocarcinoma. Carlumab (also known as CNTO 888) is a discontinued human IgG1κ monoclonal antibody that selectively binds and inhibits CC-chemokine ligand 2 (CCL2, also known as monocyte chemoattractant protein-1, MCP1), a chemokine involved in tumor angiogenesis, proliferation, invasion, metastasis, and recruitment of tumor-associated macrophages. Gemcitabine is a cytidine analog antineoplastic used as standard chemotherapy in various solid tumors, primarily acting as an antimetabolite that inhibits DNA synthesis. Preclinical and early clinical studies suggested the combination might have synergistic antitumor effects; however, carlumab was discontinued in development due to limited efficacy.
