Overview

Carmustine is a small molecule chemotherapy drug classified as a nitrosourea alkylating agent. It is primarily used to treat various malignancies, including brain tumors (such as glioblastoma, brainstem glioma, medulloblastoma, astrocytoma, ependymoma, and metastatic brain tumors), multiple myeloma (in combination with prednisone), Hodgkin's lymphoma, and non-Hodgkin's lymphoma. Carmustine works by forming interstrand crosslinks in DNA and RNA through alkylation and carbamoylation of proteins such as glutathione reductase. This disrupts DNA replication and transcription processes in cancer cells, leading to cell death. Carmustine can be administered intravenously or as an implantable wafer (Gliadel) during surgery for certain brain tumors[1][2][3][4][5][6].
