Carotuximab is a chimeric (human/murine) monoclonal antibody that targets endoglin (CD105), a glycoprotein highly expressed on proliferating endothelial cells and required for angiogenesis. By binding to CD105, carotuximab inhibits tumor angiogenesis and cell proliferation, exerting antiangiogenic and antineoplastic effects. It has been investigated as monotherapy or in combination with chemotherapy or anti–VEGF agents in various cancers and eye disorders. Carotuximab was originally developed by Tracon Pharmaceuticals, with further development by Enviro Therapeutics, Santen Pharmaceutical, the National Cancer Institute (USA), University of Alabama at Birmingham, and University of Minnesota. Its primary indications have included solid tumors such as angiosarcoma (Phase III), prostate cancer (Phase II), non-small cell lung cancer (Phase I), as well as persistent exudative age-related macular degeneration[1][2][3][4][5].
