Carrier-platin is a novel macromolecular platinum-based chemotherapeutic agent developed to overcome the limitations of small-molecule platinum drugs like cisplatin and oxaliplatin. It consists of L-aspartic acid-chelated platinum moieties (Asp-Pt) conjugated to a poly(L-glutamic acid/L-aspartic acid) backbone. This poly(amino acid) conjugation strategy is designed to improve pharmacokinetics, increase tumor accumulation through the enhanced permeability and retention (EPR) effect, and bypass multi-drug resistance (MDR) proteins that often lead to treatment failure. In preclinical studies, carrier-platin demonstrated potent anti-tumor efficacy in colorectal and ovarian cancer models, including those resistant to cisplatin and oxaliplatin, while maintaining a superior safety profile with reduced hematotoxicity and weight loss compared to standard platinum therapies.
