Overview

CART-19 (Chinese PLA General Hospital) is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy designed to target the CD19 antigen, which is ubiquitously expressed on B-cell malignancies. Developed by the Chinese PLA General Hospital, this therapy is specifically being evaluated for the treatment of relapsed or refractory mantle cell lymphoma (MCL), particularly in elderly patients who have limited treatment options. The CAR construct is a second-generation design that incorporates a CD19-specific single-chain variable fragment (scFv) coupled with the CD137 (4-1BB) costimulatory domain and the CD3-zeta signaling domain. The treatment process involves the ex vivo genetic modification of a patient's own T cells using a viral vector to express the CAR, followed by the re-infusion of these engineered cells into the patient to induce targeted cytotoxicity against CD19-positive lymphoma cells.
