Overview

CART-19 cell (CD19-directed chimeric antigen receptor T-cell) therapy is an autologous cell therapy designed to target and eliminate CD19-expressing B-cell malignancies. The therapy involves isolating a patient's T-cells, genetically engineering them using a viral vector (typically lentivirus or retrovirus) to express a chimeric antigen receptor (CAR) specific for CD19, and reinfusing them back into the patient. The CAR construct typically consists of an anti-CD19 single-chain variable fragment (scFv) linked to a costimulatory domain (such as 4-1BB/CD137 or CD28) and the CD3ζ signaling domain. CART-19 cell therapy is widely investigated and approved for various B-cell malignancies, including B-cell acute lymphoblastic leukemia (B-ALL), chronic lymphocytic leukemia (CLL), and various B-cell lymphomas. In China, Luoyang Central Hospital, in collaboration with Henan Huarong Biotechnology, has evaluated CART-19 cell therapy in clinical trials (such as ChiCTR1800018211) for relapsed or refractory B-cell leukemia and lymphoma.
