CART-38 is an experimental chimeric antigen receptor (CAR) T-cell therapy targeting the CD38 antigen, developed by researchers at the University of Pennsylvania and the Children's Hospital of Philadelphia. It is designed as a pan-hematologic treatment for multiple myeloma (MM), T-cell acute lymphoblastic leukemia (T-ALL), and acute myeloid leukemia (AML). The therapy involves engineering a patient's T cells to express a CAR that recognizes CD38, a glycoprotein overexpressed on various hematologic malignancies. Preclinical data published in 2023 demonstrated that CART-38 effectively eliminates malignant cell lines and primary patient samples in xenograft models. However, the therapy also targets normal hematopoietic progenitor cells, necessitating careful clinical monitoring for potential myeloablation. A Phase 1 clinical trial is investigating its safety and efficacy in patients with relapsed or refractory multiple myeloma.
