Overview

CART-BCMA is a chimeric antigen receptor T-cell (CAR-T) therapy targeting B-cell maturation antigen (BCMA) for the treatment of multiple myeloma. It is designed with tandem T-cell receptor (TCR) and 4-1BB costimulatory domains produced by a lentiviral system. The therapy works by genetically modifying a patient's own T cells to express a receptor that recognizes BCMA, a protein highly expressed on multiple myeloma cells. Once infused back into the patient, these modified T cells can recognize and attack cancer cells expressing BCMA. The mechanism of action involves binding to BCMA on tumor cell surfaces, which triggers T-cell activation, proliferation, and cytokine release, ultimately leading to tumor cell death through various pathways including perforin and granzyme secretion.
