Overview

CART-BCMA + huCART19 is an investigational combination autologous cellular immunotherapy being developed by the University of Pennsylvania in collaboration with Novartis. This dual-targeting approach consists of two distinct CAR T-cell products: CART-BCMA, which targets B-cell maturation antigen (BCMA), and huCART19 (also known as CTL119), which targets CD19. Both products are autologous T cells engineered via lentiviral transduction to express chimeric antigen receptors (CARs) containing 4-1BB and CD3ζ signaling domains. By targeting both BCMA and CD19, the therapy aims to address the heterogeneity of antigen expression on malignant plasma cells and potentially eliminate myeloma progenitor cells. The combination is currently being evaluated in a Phase 1 clinical trial (NCT03549442) for patients with high-risk multiple myeloma, either as a consolidation strategy following first- or second-line therapy or for relapsed/refractory disease responding to salvage therapy.
