CART-EGFR-IL13Rα2-dnTGFβRII is an experimental tri-modular chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of glioblastoma (GBM). The therapy utilizes a bicistronic construct that simultaneously targets two tumor-associated antigens: the Epidermal Growth Factor Receptor (EGFR), using the 806 scFv which targets a specific tumor-restricted conformation, and the Interleukin-13 Receptor Alpha 2 (IL13Rα2), using the Hu07 scFv. To combat the immunosuppressive tumor microenvironment characteristic of GBM, the T cells are further engineered to express a dominant-negative TGF-beta receptor II (dnTGFβRII). This truncated receptor lacks the intracellular signaling domain, allowing it to sequester extracellular TGF-β (acting as a "sink") and prevent TGF-β-mediated suppression of the CAR-T cells' proliferation and effector functions. Developed at the University of Pennsylvania, this construct is designed to overcome antigen heterogeneity and immune evasion in malignant gliomas.
