CART-EpCAM is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy targeting the Epithelial Cell Adhesion Molecule (EpCAM), a transmembrane glycoprotein highly expressed in various epithelial-derived solid tumors. Developed by investigator Su Haichuan, the therapy involves genetically modifying a patient's own T cells to express a CAR specific to EpCAM, enabling them to recognize and eliminate cancer cells in indications such as pancreatic, colorectal, gastric, and lung cancers. To enhance clinical safety, the CAR construct incorporates an epidermal growth factor receptor (EGFR)-based safety switch, which allows for the targeted depletion of the engineered cells in the event of severe adverse effects. The therapy is currently undergoing Phase 1 clinical evaluation (NCT04151186) to determine its safety, tolerability, and preliminary anti-tumor activity.
