Overview

CART-meso-19 is an investigational **co-administration regimen of two separately manufactured autologous lentivirally transduced CAR-T-cell products** developed at the University of Pennsylvania for metastatic pancreatic cancer. CART-meso cells express an anti-mesothelin CAR incorporating an SS1-derived single-chain variable fragment with 4-1BB and CD3-zeta signaling domains to direct cytotoxic T cells against mesothelin-expressing tumor cells. CART19 cells express a humanized anti-CD19 CAR with 4-1BB and CD3-zeta signaling domains and are intended to deplete CD19-positive B cells, thereby reducing anti-CAR humoral responses and potentially improving persistence of CART-meso cells. In the Phase 1 NCT02465983 study, the two cell products were infused intravenously as separate infusions after cyclophosphamide lymphodepletion; the trial was terminated after enrolling four participants.
