CART-PSMA-TGFβRDN is an autologous chimeric antigen receptor (CAR) T-cell therapy designed to treat metastatic castration-resistant prostate cancer (mCRPC). The T cells are genetically modified using a lentiviral vector to express a CAR targeting Prostate-Specific Membrane Antigen (PSMA). A key feature of this specific construct is the co-expression of a dominant-negative TGF-beta type II receptor (TGFβRDN). This modification is intended to make the CAR-T cells resistant to the immunosuppressive effects of Transforming Growth Factor-beta (TGF-β) within the tumor microenvironment, thereby enhancing T-cell persistence and anti-tumor activity. It was originally developed at the University of Pennsylvania and is being advanced by Tceleron Therapeutics.
