Overview

CART-TnMUC1 is an investigational chimeric antigen receptor T-cell (CAR-T) therapy designed to target the tumor-associated glycoform TnMUC1, an aberrant form of the mucin 1 (MUC1) protein commonly expressed on a variety of solid tumors and multiple myeloma. The therapy involves genetically modifying autologous T cells to express a CAR that specifically recognizes TnMUC1, enabling targeted immune-mediated destruction of cancer cells. The CAR construct includes a CD2 co-stimulatory domain to enhance T cell activation and anti-tumor response. This approach aims to overcome challenges associated with treating solid tumors using CAR-T therapies, which have previously shown limited efficacy in this setting compared to hematologic malignancies. Primary indications under investigation include advanced solid tumors such as ovarian cancer, pancreatic ductal adenocarcinoma, triple-negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), and multiple myeloma.
