Overview

CART001 is an investigational switchable chimeric antigen receptor (CAR) T-cell therapy designed to recognize fluorescein isothiocyanate (FITC). Developed by the National Cancer Center Japan and Yamaguchi University, it is part of a binary system where the CAR-T cells are activated only in the presence of a FITC-labeled adapter molecule (the switch). In clinical studies for CD20-positive B-cell non-Hodgkin lymphoma, it is used in conjunction with FY001, a FITC-labeled rituximab. This approach allows for the modulation of CAR-T activity by controlling the dose and timing of the adapter molecule, potentially improving safety by reducing the risk of cytokine release syndrome (CRS) and neurotoxicity, and allowing for multi-antigen targeting by changing the adapter.
