Overview

CART19-BE-01 is an autologous chimeric antigen receptor (CAR) T-cell therapy developed by the Fundación de Investigación Biomédica de Salamanca. The patient's T-cells are genetically modified using a lentiviral vector to express a CAR targeting the CD19 antigen, which is widely expressed on B-cell malignancies. The CAR construct incorporates a 4-1BB (CD137) costimulatory domain and a CD3-zeta signaling domain to enhance T-cell persistence and anti-tumor activity. Additionally, the cells express a truncated human epidermal growth factor receptor (huEGFRt), which serves as a selection marker during manufacturing and a potential safety switch for antibody-mediated ablation using cetuximab. It is currently being evaluated in Phase 2 clinical trials for various relapsed or refractory B-cell lymphomas, including diffuse large B-cell lymphoma (DLBCL) and mantle cell lymphoma (MCL).
