Overview

CART19-GFP is a preclinical-stage, non-activated chimeric antigen receptor (CAR) T-cell therapy targeting CD19, used primarily as a control construct in the development of cytokine-armed CAR-T therapies. Developed by researchers at the University of Pennsylvania, it is manufactured using a rapid, 1-day protocol that bypasses traditional T-cell activation and expansion steps. This approach aims to preserve a na\u00efve-like T-cell phenotype, which is associated with enhanced persistence and reduced exhaustion. In preclinical studies, CART19-GFP serves as the baseline comparator for IL-18-secreting CAR-T cells (CART19-IL18) in models of B-cell leukemia and lymphoma. The construct co-expresses Green Fluorescent Protein (GFP) as a reporter to facilitate tracking and phenotypic analysis.
