Overview

CART22 is an autologous chimeric antigen receptor (CAR) T-cell therapy designed to target the CD22 antigen found on B-cell malignancies. It involves genetically modifying a patient's own T cells to express a CAR that recognizes CD22. Specifically, the CART22-65s construct, a fully human anti-CD22/4-1BB CAR T-cell, incorporates tandem TCR-ζ and 4-1BB co-stimulatory signaling domains to enhance T-cell activation and persistence. This cell therapy is being developed for the treatment of relapsed or refractory B-cell Acute Lymphoblastic Leukemia (B-ALL), particularly in patients who have relapsed after prior CD19-directed CAR T-cell therapy, addressing mechanisms of relapse such as CD19-antigen escape. It aims to redirect the patient's T cells to identify and eliminate CD22-expressing cancer cells.
