Overview

CART33Arg is an experimental allele-specific chimeric antigen receptor (CAR) T-cell therapy designed to target a specific single nucleotide polymorphism (SNP) in the CD33 (SIGLEC3) gene. Specifically, it targets the Arg69 variant (rs2455069) of CD33. This approach is intended for patients with acute myeloid leukemia (AML) who have relapsed after an allogeneic hematopoietic cell transplant (HCT). By targeting an allele present on the recipient's leukemia cells but absent on the donor-derived healthy myeloid cells (which would carry the alternative Gly69 allele), CART33Arg aims to provide potent anti-leukemic activity while selectively sparing the donor's hematopoietic system from on-target, off-tumor toxicity. It was developed by researchers at Washington University School of Medicine.
