CART33Gly is an allele-specific chimeric antigen receptor (CAR) T-cell therapy designed to target a specific single nucleotide polymorphism (SNP) in the CD33 (SIGLEC3) gene. Specifically, it targets the Gly69 variant (rs2455069) of CD33. This approach is intended for patients with relapsed acute myeloid leukemia (AML) following allogeneic hematopoietic cell transplantation (HCT). By targeting the recipient's leukemia cells expressing the Gly69 allele while selectively sparing donor-derived hematopoietic cells that express the alternative Arg69 allele, the therapy aims to reduce on-target, off-tumor toxicity (myelosuppression) typically associated with pan-CD33 CAR-T therapies. It was developed by researchers at Washington University School of Medicine.
