Overview

CART33Pan is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target a pan-epitope of the CD33 (SIGLEC3) antigen, which is highly expressed on acute myeloid leukemia (AML) cells. Developed by researchers at Washington University School of Medicine, it serves as a benchmark or positive control in studies evaluating the efficacy and specificity of allele-specific CAR-T cells (such as CART33Gly and CART33Arg). While allele-specific CARs are designed to exploit single nucleotide polymorphisms (SNPs) like rs2455069 to selectively spare donor-derived hematopoietic cells post-allogeneic transplant, CART33Pan targets CD33 broadly across different polymorphisms. Preclinical in vitro studies have demonstrated that CART33Pan exhibits potent cytotoxicity against various CD33-expressing AML cell lines, regardless of their specific CD33 allele status.
