CART45RA-NKG2D is an investigational allogeneic CAR-T cell therapy developed by the Fundación para la Investigación Biomédica del Hospital Universitario La Paz. It consists of T cells derived from healthy donors that have been depleted of CD45RA+ (naive) T cells to minimize the risk of graft-versus-host disease (GvHD) and then engineered to express a chimeric antigen receptor (CAR) based on the NKG2D receptor. The NKG2D CAR targets NKG2D ligands (such as MICA, MICB, and ULBP1-6), which are frequently upregulated on the surface of various hematological malignancies, including B-cell and T-cell acute lymphoblastic leukemia (ALL). By utilizing the natural binding properties of the NKG2D receptor, this therapy aims to provide a broad-spectrum anti-leukemic effect across different subtypes of refractory or relapsed ALL.
