Overview

CART72 is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy developed for the treatment of metastatic colorectal cancer. The therapy involves engineering a patient's own T cells to express a CAR targeting tumor-associated glycoprotein 72 (TAG-72), a cell surface antigen commonly expressed in colorectal and other epithelial cancers. The CAR construct in CART72 uses a humanized version of the murine CC49 antibody specific for TAG-72, fused with a CD3-zeta intracellular signaling domain to activate T cells upon antigen binding. In early phase 1 clinical trials, CART72 was administered either intravenously or via hepatic artery infusion, sometimes alongside interferon-alpha to upregulate TAG-72 expression on tumor cells. While the manufacturing success rate was high and some trafficking of infused cells to tumors was observed, persistence in patients' blood was generally short-lived (≤14 weeks), likely due to immunogenicity against murine components of the CAR construct. Low-grade cytokine release syndrome occurred but no severe on-target/off-tumor toxicity was reported[1][2][7].
