Overview

CARTemis-1-28 is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting B-cell maturation antigen (BCMA), specifically engineered with a CD28 costimulatory domain. Developed by researchers at the Instituto de Biomedicina de Sevilla, it is designed for the treatment of multiple myeloma. The product is manufactured using an 8-day process involving activation with anti-CD3/CD28 beads and expansion in the presence of IL-7 and IL-15. In comparative studies with its 4-1BB-containing counterpart (CARTemis-1-BB), CARTemis-1-28 demonstrated similar initial cytotoxicity but showed lower persistence and reduced mitochondrial respiratory capacity following repeated antigen stimulation, potentially due to overexpression of genes in the TGF-β signaling pathway.
