Overview

CARv3-TEAM-E T cells are an investigational chimeric antigen receptor (CAR) T cell therapy designed for the treatment of glioblastoma. This autologous cell therapy is engineered to target both the epidermal growth factor receptor variant III (EGFRvIII), a tumor-specific antigen, and wild-type EGFR protein through two mechanisms. The first is a second-generation anti-EGFRvIII CAR with an intracellular 4-1BB costimulatory domain linked to CD3 zeta; the second involves secretion of a T-cell–engaging antibody molecule (TEAM) against wild-type EGFR, which is highly expressed in glioblastoma but not in normal brain tissue. The dual-targeting approach aims to address tumor heterogeneity and enhance antitumor activity by redirecting even regulatory T cells against tumors at the site of antigen engagement. Developed at Massachusetts General Hospital, this product has shown dramatic but sometimes transient radiographic tumor regression in early-phase clinical trials for recurrent or newly diagnosed glioblastoma[1][4][5][6][7].
