Carvedilol + amlodipine + olmesartan is a hypothetical combination drug comprised of three antihypertensive agents: *carvedilol* (a non-selective beta-adrenergic and alpha-1 blocker), *amlodipine* (a calcium channel blocker), and *olmesartan* (an angiotensin II receptor blocker). Each component lowers blood pressure through a distinct mechanism: Carvedilol blocks beta and alpha-1 adrenergic receptors, reducing heart rate and vascular resistance; amlodipine inhibits L-type calcium channels in vascular smooth muscle, causing vasodilation; olmesartan blocks angiotensin II type 1 (AT1) receptors, preventing vasoconstriction and aldosterone release. The combination could offer additive antihypertensive effects and may be considered for patients needing multi-mechanistic blood pressure control. There is no currently marketed fixed-dose combination containing all three components; such a product would represent an extension of existing combination therapies currently used for difficult-to-control hypertension[1][2][5][7].
