Casdozokitug is a first-in-class human immunoglobulin G1 (IgG1) monoclonal antibody that targets the p28 subunit of interleukin-27 (IL-27p28), an immunosuppressive cytokine. By inhibiting IL-27 activity, casdozokitug blocks its immune-suppressing effects in the tumor microenvironment and promotes immune cell activation. This mechanism enables anti-tumor activity and may help overcome resistance to checkpoint inhibitors in certain cancers. Casdozokitug is being developed primarily for solid tumors such as hepatocellular carcinoma (HCC), non-small cell lung cancer (NSCLC), and renal cell carcinoma (RCC). It has received Orphan Drug designation and Fast Track designation from the FDA for refractory HCC. The drug was originally developed by Surface Oncology and is now being advanced by Coherus BioSciences following their acquisition of Surface Oncology.
