Overview

Casein phosphopeptide (CPP) is a phosphorylated peptide derived from the enzymatic hydrolysis of casein, the main protein in milk. It contains clusters of phosphorylated serine residues that enable it to chelate and stabilize calcium and phosphate ions in solution[5][6][8]. This property allows CPP to act as a carrier for bioavailable minerals—especially calcium—preventing their precipitation and enhancing their intestinal absorption[5][10]. In dental applications, CPP is often combined with amorphous calcium phosphate (ACP) to form CPP-ACP complexes that help buffer free calcium and phosphate ion activities in dental plaque. This maintains supersaturation with respect to tooth enamel, reduces demineralization, promotes remineralization of subsurface lesions (such as white spot lesions), inhibits cariogenic activity, and protects against tooth sensitivity and erosion[2][7]. As a dietary supplement or functional food ingredient, casein phosphopeptides are also studied for their potential roles in improving mineral absorption (notably calcium), lowering blood pressure via vasodilation effects (as certain lactotripeptides), reducing risk of eczema or allergies when used in infant formulas for at-risk populations[1], and possibly supporting bone health by increasing serum osteocalcin levels while decreasing markers like alkaline phosphatase[5].
