Casp4i is a discovery-stage small peptidomimetic inhibitor of caspase-4, developed by IPCURE. Caspase-4 is an essential component of the non-canonical inflammasome pathway, acting as an intracellular receptor for bacterial lipopolysaccharide (LPS). Upon activation, caspase-4 cleaves gasdermin D (GSDMD), leading to the formation of membrane pores that induce pyroptotic cell death and the unconventional secretion of pro-inflammatory cytokines such as IL-1β and IL-18. By targeting this pathway, IPCURE aims to address various inflammatory conditions where caspase-4-mediated pyroptosis contributes to disease pathology. The compound is part of a specialized pipeline of peptidomimetics designed to provide high specificity and improved pharmacological properties compared to traditional peptide-based caspase inhibitors.
