Overview

Caspofungin + liposomal amphotericin B is a combination antifungal therapy used primarily for the treatment of severe invasive fungal infections, particularly in immunocompromised patients. Caspofungin is an echinocandin that inhibits the synthesis of β-(1,3)-D-glucan, an essential component of the fungal cell wall. Liposomal amphotericin B is a polyene antifungal that binds to ergosterol in fungal cell membranes, creating pores and causing cell death. The combination targets different components of the fungal cell structure and has been studied for synergistic or additive effects against pathogens such as Aspergillus spp., Candida spp., and Mucorales. Clinical studies suggest this regimen may improve outcomes compared to monotherapy in certain high-risk populations, with acceptable safety profiles[1][5][6][7].
