caStx1 is a chimeric monoclonal antibody specifically engineered to target and neutralize Shiga toxin 1 (Stx1), a potent cytotoxin produced by Shiga toxin-producing *Escherichia coli* (STEC). Developed by Thallion Pharmaceuticals, caStx1 was intended to be administered as part of a dual-antibody cocktail named Shigamabs, which also included caStx2. The primary therapeutic goal was to prevent the progression of STEC infections to Hemolytic Uremic Syndrome (HUS), a life-threatening condition characterized by acute renal failure, hemolytic anemia, and thrombocytopenia. The antibody functions by binding to the Stx1 toxin, thereby preventing its attachment to the globotriaosylceramide (Gb3) receptors on the surface of target cells, effectively neutralizing the toxin's ability to inhibit protein synthesis and cause cell death. Clinical development reached Phase 2 trials before being discontinued following corporate restructuring and the acquisition of Thallion by Bellus Health.
