CAT-101 is an autologous dendritic cell-based cancer vaccine consisting of engineered dendritic cells (eDCs) loaded with mutant KRAS (mKRAS) antigens. Developed by the Cancer Institute and Hospital, Chinese Academy of Medical Sciences, it is designed to stimulate a T-cell-mediated immune response against tumor cells harboring specific KRAS mutations, including G12C, G12D, G12R, and G12V. The therapy is specifically being evaluated in HLA-A*1101-positive patients with advanced solid tumors, particularly pancreatic cancer, either as a monotherapy or in combination with the PD-1 inhibitor tislelizumab. By presenting these specific mutant antigens, CAT-101 aims to activate mutation-specific T cells to recognize and eliminate cancer cells while sparing healthy tissue.
