Overview

CAT-2054 is an investigational oral small molecule drug developed by Catabasis Pharmaceuticals. It acts as an inhibitor of Sterol Regulatory Element-Binding Protein (SREBP), a master regulator of lipid metabolism. By modulating SREBP activity, CAT-2054 may inhibit the production of key cholesterol metabolism proteins such as PCSK9, HMG-CoA reductase, ATP citrate lyase, and NPC1L1. This mechanism has the potential to impact LDL cholesterol, triglycerides, glucose metabolism, and liver fat. Initially developed for hypercholesterolemia in patients inadequately managed by existing therapies, it has also been investigated for nonalcoholic steatohepatitis (NASH). In preclinical models and early clinical trials (Phase 1 and Phase 2a), CAT-2054 showed potential benefits on lipid parameters and liver health; however, in a Phase 2a trial with high-intensity statin therapy in hypercholesterolemia patients, no effect on LDL-C was observed[1][3][5][6].
