CAT-248 is an allogeneic, off-the-shelf CAR-NK cell therapy developed by Catamaran Bio for the treatment of CD70-expressing solid tumors. The product consists of healthy donor-derived peripheral blood natural killer (NK) cells that are engineered to express a chimeric antigen receptor (CAR) targeting CD70, as well as interleukin 15 (IL-15) and a dominant negative TGFβ receptor. Additionally, the endogenous CD70 locus is knocked out in these NK cells. CAT-248 is manufactured using non-viral TcBuster transposon technology and can be produced at clinical scale within 15 days. Preclinical studies have demonstrated that CAT-248 exhibits potent cytotoxicity against cancer cell lines with varying levels of CD70 expression and shows durable anti-tumor activity in xenograft models of renal cell carcinoma (RCC) and non-small cell lung cancer (NSCLC). The therapy expands and persists in vivo after intravenous administration, effectively infiltrating tumors and eliminating CD70-positive cancer cells[1][2][3].
