CAT-ApMnkQ2 is an RNA aptamer candidate developed by Aptus Biotech for the treatment of anaplastic thyroid cancer (CAT). It is an optimized RNA version of apMNKQ2, a DNA aptamer that targets mitogen-activated protein kinase-interacting serine/threonine-protein kinase 1 (MNK1), specifically the MNK1b isoform. By binding to MNK1, CAT-ApMnkQ2 inhibits the MNK-eIF4E signaling pathway, which is overexpressed in various cancers and plays a critical role in tumor progression, metastasis, and cancer stem cell maintenance. Preclinical studies have demonstrated that targeting this pathway with CAT-ApMnkQ2 induces apoptosis, arrests cell cycle progression, and reduces tumor growth, migration, and invasion, making it a promising therapeutic strategy for highly aggressive and chemotherapy-resistant cancers like anaplastic thyroid cancer.
