Cat-SPIRE is a synthetic peptide immunotherapy vaccine candidate developed for the treatment of cat allergy. It utilizes Synthetic Peptide Immuno-Regulatory Epitopes (Spires), which are short amino acid sequences designed to induce immune tolerance specifically to the major cat allergen, Fel d 1. The approach aims to modulate the immune response and reduce allergic symptoms by targeting T-cell responses rather than relying on traditional whole-protein or extract-based immunotherapies. Cat-SPIRE was developed by Circassia in collaboration with researchers at Imperial College London. Despite promising early clinical results, a phase 3 trial failed due to an unexpectedly strong placebo effect, leading to discontinuation of its development[2].
