Overview

Cathepsin E short hairpin RNA is an RNA interference (RNAi) based therapeutic agent designed to specifically silence the expression of the *CTSE* gene. Cathepsin E is an intracellular aspartic protease that plays a critical role in colorectal cancer (CRC) progression by promoting metastasis and facilitating immune evasion. The mechanism involves the ADP-ribosylation of ZNF326 and the subsequent activation of the TGF-β/SMAD signaling pathway, which drives tumor growth and suppresses anti-tumor immune responses. By delivering shRNA sequences—often via viral vectors or lipid nanoparticles—the agent triggers the sequence-specific degradation of CTSE mRNA, thereby reducing protein levels and potentially inhibiting the malignant phenotype of CRC cells.
