Overview

This cathepsin S inhibitor is a small molecule therapeutic candidate being developed by The First Hospital of Guangxi Medical University for the treatment of liver cancer, specifically hepatocellular carcinoma (HCC). Cathepsin S (CTSS) is a lysosomal cysteine protease that is frequently overexpressed in various malignancies, where it plays a critical role in tumor progression, angiogenesis, and metastasis by degrading extracellular matrix components and modulating the tumor microenvironment. In the context of liver cancer, research from Guangxi Medical University has demonstrated that cathepsin S promotes tumor cell proliferation, invasion, and the epithelial-mesenchymal transition (EMT). Inhibition of this enzyme has been shown to suppress these oncogenic processes and may also influence autophagy and immune evasion mechanisms, such as M2 macrophage polarization, within the liver tumor niche. The program is currently in the early stages of development, focusing on the potential of CTSS inhibition to treat advanced or metastatic liver malignancies.
