**CAV2-AU-M3** is a preclinical, conditionally replicative canine oncolytic adenovirus developed by Auburn University for canine osteosarcoma. The engineered virus infects and lyses canine osteosarcoma cells while encoding a secreted anti-programmed cell death protein 1 heavy-chain antibody. Local antibody production in the tumor microenvironment is intended to block the programmed cell death protein 1 and programmed death-ligand 1 immune-checkpoint interaction, complementing direct virus-mediated tumor cell lysis and promoting antitumor immunity. Preclinical studies evaluated activity in canine osteosarcoma cell lines, three-dimensional spheroids, and an intratumoral mouse tumor model.
