Drug intelligence / Profile preview
Caveolin-1 peptide refers to a class of investigational synthetic peptides derived from the caveolin-1 scaffolding domain (CSD, amino acids 82–101 of caveolin-1). Caveolin-1 (Cav-1) is a membrane protein essential for tissue homeostasis, but its expression is significantly downregulated in fibrotic diseases such as idiopathic pulmonary fibrosis (IPF). Caveolin-1 peptides act as functional mimetics or replacements for endogenous Cav-1, binding to proteins containing caveolin-binding domains (CBD) to regulate cellular signaling. The lead clinical candidate in this class, LTI-03 (also known as CSP7), is a 7-amino acid peptide formulated as an excipient-free dry powder for inhalation. It exerts a dual mechanism of action: it attenuates pro-fibrotic signaling pathways (such as TGF-β/Smad, PKC, and MAPK) to reduce lung scarring, while simultaneously promoting the survival and regeneration of alveolar epithelial cells by inhibiting p53-mediated apoptosis and senescence. LTI-03 is currently in Phase 2 clinical development for the treatment of IPF.
