Cavrotolimod is a synthetic spherical nucleic acid (SNA) designed as an agonist of Toll-like receptor 9 (TLR9). It acts as a potent innate immune activator, specifically stimulating Th1-type immune responses by targeting and activating TLR9 within endosomes of various immune cells, including monocytes/macrophages, plasmacytoid dendritic cells (pDCs), natural killer (NK) cells, and B cells. Cavrotolimod is being developed primarily for the treatment of solid tumors such as advanced Merkel cell carcinoma (MCC), cutaneous squamous cell carcinoma (CSCC), and chronic hepatitis B virus infection. The drug has demonstrated anti-tumor effects in preclinical models and early-phase clinical trials by inducing cytokine production, enhancing antigen-presenting cell activity, and promoting both innate and adaptive immune responses. Cavrotolimod was originally developed by Exicure but is now being advanced by Bluejay Therapeutics.
